I've tried a few solutions that work OK (including Groove), but in the end when it comes to flexibility I have all my stuff on a personal server that I can access from anywhere, and A computer in my server room that I can login to from anywhere (logmein.com - Free account) and access any large files or applications instantly.
As far as emails, I use Gmail with iMAP on my phone, Laptop and other computers then not matter what you do or change it is automatically updated across all your devices.
I suggest "Postbox" for your any of your PC, or MAC computers it has gmails iMAP settings built in, just type in username and password.

Hope this helps a little, I know its not a all in one solution but I just have not found anything better then these combos.
